I've been doing reseach about chitinase activity from hot spring's bacteria..at first,for the enzyme assay I use shcale method (colorimetric). this method measures the product of enzyme (N-acetylgukosamin)during incubation. but I've failed many times..so I was looking for another method..until I finally found turbidimetric method. this method measures residual substrate from sample during incubation.
if using a schale method,the number of units is only around 0,... but if using a turbidimetry method, the number of units could reach 4,...
my questions:
-what is definition of Unit/ml enzyme? is it different for each method?
- is turbidimetric method accurate for chitinase assay?
