U.S. Route 27 enters Indiana
in southeastern part of the state south of the village of Liberty. At Liberty, US 27 turns north, paralleling the Ohio state line. It intersects Interstate 70
/U.S. Route 40 (I-70/US 40) at Richmond and continues north to Decatur. US 33
joins US 27 just south of Decatur and the two routes share a northwesterly alignment from Decatur to I-469/US 24
south of Fort Wayne. US 33 turns west and follows I-469 and US 24, while US 27 continues north into downtown Fort Wayne were northbound US 27 is on Spy Run Avenue and southbound is on Clinton Street. North of downtown, US 27 follows Lima Road to its terminus at I-69

In November 1967, Indiana completed Interstate 69, the last incomplete section being opened between Angola
and the Michigan/Indiana state line, supplanting the old two-lane surface road as a through route. Interstate 69 north of Fort Wayne had been completed in stages, the designation of US 27 being moved to the new freeway. Some segments of old US 27 remained as state highways with designations as SR 327, SR 427, and SR 127. Parts of these routes, comprising what had been had been the old surface road, were eventually returned to country governments. Within greater Fort Wayne, US 27 was eventually diverted from Coldwater Road to what had been Indiana 3 (SR 3/Lima Road).
In 2001, Indiana removed the US 27 designation north of its interchange with I-69 as superfluous. Some rare signs are visible along I-69 on overhead signs as far north as Auburn

On March 9, 2007, legislation was introduced in the Indiana House of Representatives
to designate US 27 as historic highway. Richmond, one of the cities through which US 27 passes, would have two historic highways passing through it as the original National Road (U.S. Route 40
) runs through Richmond as well.
