The 2010–11 season is the 105th season of the Isthmian League
Premier Division and the 5th season for the current incarnations of the Division One North and Division One South. It runs from August 2010 to May 2011.

In the First Round, the four lowest ranked clubs in the Isthmian League played each other for a place in the Second Round.
The two clubs to have made it through the First Round were entered into the draw with every other Isthmian League club, making sixty-four teams.
